How the calculation works

Concentration is the mass of peptide divided by the volume of solvent it is dissolved in:

concentration (mg/ml) = peptide mass (mg) ÷ solvent volume (ml)

Rearranged, that also tells you how much solvent to add to reach a concentration you're aiming for:

solvent volume (ml) = peptide mass (mg) ÷ target concentration (mg/ml)

Adding less solvent gives a more concentrated solution; adding more gives a more dilute one. The quantity of peptide in the vial never changes — only the concentration of the solution you prepare from it.
